SuR RNA-seq Library Prep Kit for Illumina® is suitable for the library construction of a strand-specific transcriptome with a starting template of 0.1-1 μg total RNA from human, mouse or rat. Unlike conventional mRNA-seq library construction, this kit removes rRNA (including cytoplasmic 28S, 18S, 5S rRNA and mitochondrial 12S, 5.8S rRNA) from total RNA, retaining mRNA and other non-coding RNAs, and can be used for long non-coding (lncRNA) analysis.
dUTP is labeled in the synthesis of second strand cDNA. The labeled second strand template is digested with uracil-DNA glycosylase (UDG) before PCR, so that the final sequencing information is from the first strand cDNA, thereby retaining the strand orientation of mRNA. Data analysis after sequencing can determine whether the transcript is from a sense or antisense DNA strand.
Degraded RNA samples can also be used for library construction using this kit.
