SuR-sc Single Cell Kit V2 is suitable for non-bias Phi29 isothermal genome amplification system based on multiple displacement amplification (MDA). The Phi 29 DNA polymerase used in this kit is a DNA polymerase from phage. This polymerase has strong strand displacement activity and can realize the melting and replication of DNA with complex structures. Phi 29 DNA polymerase also has a strong chain affinity. In a single polymerization reaction, continuous polymerization extension can reach up to 100 kb and its amplification product is suitable for almost all downstream applications. Phi 29 DNA polymerase also has strong 3'-5' exonuclease activity. The fidelity is 1000 times that of the Taq polymerase, which guarantees the high fidelity of DNA synthesis.
This kit uses the optimized Phi29 DNA polymerase reaction system to achieve highly sensitive single-cell whole-genome isothermal amplification. It is suitable for whole genome amplification from single cell, small amount of tissue or micro-purified genomic DNA to obtain a large amount of high coverage genomic DNA.
