SuR-sc WTA Kit V2 can use 1-1000 cells or 10 pg-10 ng total RNA as templates for first-strand cDNA synthesis and amplification to obtain enough samples for sequence analysis, which are technical challenges by conventional mRNA-seq.

The kit uses Oligo dT primer as a reverse transcription primer for cDNA synthesis, and uses the template-switching activity of reverse transcriptase to add a linker sequence at the 3' end of the cDNA. Subsequent PCR is performed to obtain the full-length cDNA. This process can effectively avoid 3' preference and rRNA contamination during cDNA synthesis.
