Abstract

RNA-based therapeutics are transforming medicine, but scaling production efficiently remains a challenge, particularly the cost-intensive in vitro transcription (IVT) step.

Join this webinar to discover how fed-batch IVT, a strategy involving gradual reagent addition, can significantly improve mRNA yield and reagent usage efficiency. Using the Beckman Coulter BioLector XT Microbioreactor and the Cytiva ReadyToProcess WAVE™ 25 rocking bioreactor, Cytiva’s application specialist will demonstrate how to streamline process development and scale-up for fed-batch mRNA synthesis.

Learn how microfluidic feeding and real-time monitoring with the BioLector XT Microbioreactor enabled rapid optimization of feeding strategies and feed stock concentrations in sub-milliliter reactions. See how optimized fed-batch IVT conditions improved mRNA yield, reaching up to 15 g/L based on the initial IVT volume, while improving NTP incorporation efficiency and reducing DNA template use.

You’ll also discover how pH showed a linear correlation with mRNA crude concentration, and could be leveraged as a non-invasive, real-time indicator of mRNA synthesis. Finally, see how these process insights translate seamlessly to the Cytiva ReadyToProcess WAVE™ 25 rocking bioreactor, a closed, GMP-ready system, supporting IVT volumes from 20 mL to 20 L.

Learning objectives:

  1. Understand how implementing fed-batch IVT strategies can increase mRNA yield and optimize reagent use in manufacturing processes.
  2. Discover how the BioLector XT Microbioreactor enables rapid development and optimization of fed-batch feeding strategies, and how these can be successfully scaled up to GMP-ready systems such as the ReadyToProcess WAVE™ 25 rocking bioreactor.
  3. Recognize the value of real-time, non-invasive process indicators, such as pH, for monitoring mRNA crude concentration during IVT, and their impact on process control and efficiency.
