Abstract
Protein biomarker discovery, validation and utilization require technologies that deliver proteome insights with breadth and depth. Liquid chromatography mass spectrometry (LC-MS), the gold-standard of proteomics, provides unbiased protein coverage and detection of isoforms and post-translational modifications. However, achieving high sensitivity and resolution in complex, clinically relevant samples such as plasma or serum remains challenging and often requires trade-offs in throughput. In contrast, next-generation affinity-based proteomics offers exceptional sensitivity and specificity for low-abundance proteins with high-throughput in these complex matrices. This roundtable discussion will bring together key opinion leaders in proteomics to explore how integrating antibody-based methods, like proximity extension assay (PEA), with LC-MS can expand proteome coverage, facilitate biomarker discovery, and streamline the validation process, ultimately advancing clinical research.
